HPLC-SEC (High-Performance Liquid Chromatography Size-Exclusion Chromatography) will be used for size exclusion chromatography to detect protein aggregation and provide quantification and quality control for expressed proteins.
Size-exclusion chromatography separates proteins based on their size as they pass through a porous column. Larger molecules (like protein aggregates) elute first, followed by smaller molecules in order of decreasing size.
We’ll use this to identify if proteins are forming aggregates, separate aggregated proteins from monomers, and quantify the percentage of aggregated vs. non-aggregated protein.
Detect if proteins are clumping together and quantify aggregation levels over time.
Verify protein purity after expression and ensure consistent protein production.
HPLC-SEC will work alongside our existing analytical capabilities including nanoDSF for thermal stability correlation, BLI for binding activity assessment, and cell-free expression for comprehensive protein characterization.
HPLC-SEC will be particularly valuable for quality control of expressed proteins, helping ensure that proteins maintain their intended structure and don’t form aggregates that could affect experimental results.
